They do (unless it is brand new). Rotate the wheel and wiggle the chain up and down as you do so. The point where it is tightest (least vertical slack) is the tight spot. Adjust your chain with the wheel in this position.

Chain slack is usually measured with noone on the bike and the suspension unloaded.
