could be a number of things, brakes binding, partial seizure etc
check you have oil in your two stroke etc and maybe do a compression test. could be the rear brake binding as said i used to get this on my rxs it used to snatch causing the rear wheel to lock

GET THE BACK WHEEL OFF THE CROUND
PUT IT IN NEUTRAL
TURN THE BACK WHEEL
LISTEN
IT SHOULD TURN IF YOU TURN IT USING YOUR BOOT ON THE TYRE.
IF ITS A BASTARD TO TURN 1ST CHECK FOR TENSION ON THE CHAIN.
IF IT'S GEARBOX/ENGINE THEN THE CHAIN WILL GO TIGHT WHEN YOU TRY TO TURN.
TRY THE SAME TEST WITH THE BIKE IN GEAR AND THE SPARK PLUG OUT PLUG OUT.

IF THE CHAIN SEEMS OK
ITS THE WHEEL.
BRAKES MOST LIKELY BUT MAY BE BEARINGS.
PRAY IT IS THE BRAKE.

GOOD LUCK.

Does sound like a nip up (seizure) to me, although cant see why it would only happen in 4th gear unless just a coincidence.
Also check your rear pads, i had an TZR 250 reverse cylinder which i ran the rear pads down to the metal and as they heated up due to friction on the disc it would get so hot it used to weld themselves together causing my rear to lock up.

But my 1st guess would be partial seizure. Check your 2 stroke oil level and that your oil pump is working. There is a bleed screw on the oil pump, check here to see if any air in in the system. Also use a decent oil, not worth saving a £5 on a litre when a top end will cost a damn sight more.

i don't personally think it is mixture or seizing as it only happens in 4th. It could be something with the gearbox, or i have done this a couple of times myself, but in third when i down shifted too quick and locked up the back.
