if you have a shiTTy fuel pump or vacuum pump then you may need to crank the thing over a billion times before it can suck fuel from the tank, fill up the carb and then squirt it into the cyclinders.
try taking the spark plugs out and squirting a drop (and i mean a drop) of fuel into the cylinder.

Have you tried looking at your fuel cap ? There should be a small hole or pipe coming from it or elsewhere on the tank this it too allow air into the tank as the petrol is used this could create a vacuum and stop any petrol flowing to your carb. Just give it a blow too make sure its not blocked. Sometimes its just the really small things.
